چکیده

در این مطالعه به منظور بررسی تنوع ژنتیکی جمعیت‌‌های ماهی مرکب ببری در دو منطقه بندرعباس و بوشهر با استفاده از 6 جفت از نشانگرهای ملکولی میکروستلایت، تعداد 51 نمونه از مناطق بندرعباس و بوشهر جمع آوری و مقداری از بازوی (تانتاکول) هر نمونه در الکل اتیلیک 96 درصد قرارداده شد و به آزمایشگاه بیوتکنولوژی دانشگاه علوم و فنون دریایی خرمشهر منتقل گردید.DNA ژنومی به روش CTABیک درصد استخراج و کمیت وکیفیت DNA استخراجی با استفاده از روش اسپکتوفتومتر و الکتروفورز ژل آگاروز یک درصد تعیین گردید. واکنش زنجیره ای پلی مراز با استفاده از 6 جفت آغازگر ریزماهواره ای انجام گرفت و محصولات PCR روی ژل اکریل آمید 8 درصد الکتروفورز و با نیترات نقره رنگ آمیزی گردید که 4 جفت از پرایمرها پلی مورف و 2 جفت مونومورف بودند. پس از شمارش و اندازه گیری آلل ها، پارامترهای ژنتیک جمعیت با استفاده از نرم افزارهای Arlequin وGenAlex محاسبه و رابطه فیلوژنیکی بین نمونه ها با استفاده از نرم افزار TFPGA رسم گردید. نتایج بدست آمده نشان داد که میزان فاصله ژنتیکی و شباهت ژنتیکی بین مناطق به ترتیب 282/0 و 754/0 می‌باشد و میزان تمایز ژنتیکی پایین 031/0 بین مناطق مشاهده گردید. یافته های حاصل از این پژوهش نشان از تنوع ژنتیکی پایین اما معنی دار بین جمعیت های مورد مطالعه دارد و احتمالاً ماهی مرکب ببری دارای دو جمعیت می باشد. این یافته ها اطلاعات مفیدی را جهت حفاظت و مدیریت این گونه در خلیج فارس فراهم می کند.
