چکیده

کبد یکی از مهم ترین اندام های بدن مهره داران بوده که نقش مهمی در سم زدایی دارد. هدف از مطالعه ی حاضر، بررسی تاثیر نوع هضم آنزیمی، میزان درصد سرم جنین گاوی و دمای انکوباتور در کشت اولیه ی سلول های کبدی ماهی هامور معمولی (Epinephelus coioides) بود. برای انجام کار، از 5 عدد ماهی هامور معمولی استفاده شد. ابتدا بدن ماهی توسط الکل اتانول 70% ضدعفونی و پس از جدا کردن بافت کبد، توسط قیچی به قطعات ریز تقسیم شد و با استفاده از روش های متفاوت هضم آنزیمی توسط کلاژناز (تیپ 1 و 4)، تریپسین و استفاده از مواد مغذی و افزودنی در محیط کشت، سلول های آن جداسازی شدند. سپس سلول ها به مدت 2 هفته در محیط کشت Lebowitz L-15 به سه روش استفاده از هضم آنزیمی توسط تریپسین، هضم آنزیمی توسط کلاژناز (تیپ 1 و 4) و استفاده از مواد و افزودنی های مغذی، کشت داده شد. اثر شرایط متفاوت به لحاظ میزان دمای انکوباتور (20، 25، 28، 30 و32 درجه سانتیگراد) و میزان سرم جنین گاوی (0%، 10% و 20% و 20%+ITS) بر رشد سلول ها بررسی شد. براساس نتایج به دست آمده، تعداد سلول ها با استفاده از سیستم کشت هضم آنزیمی توسط آنزیم کلاژناز تیپ 4 در مقایسه با سایر روش ها بیشتر بود. در مجموع با بررسی حاضر، مناسب ترین روش برای کشت سلول های کبدی ماهی هامور معمولی، استفاده از مواد مغذی ITS، سرم جنین گاوی به میزان 20% و دمای انکوباتور C^30 تشخیص داده شده است.
